ABSTRACT

To investigate the therapeutic effects of triterpenoids from Cyclocarya paliurus on non-alcoholic fatty liver disease (NAFLD),the model of NAFLD in HepG2 cells was induced by free fatty acids (FFAs). Cytotoxicity of the triterpenoids from C. paliurus was determined by MTT method,and the effects of triterpenoids without cytotoxicity on intracellular triglyceride (TG)and superoxide dismutase (SOD)were detected by the kits. Data indicated that compound 4 [2α,3α,23-trihydroxy-12,20 (30)-dien-28-ursolic acid,TUA]had hypolipidemic and antioxidant activities. After being treated with TUA and FFAs for 24 h,the intracellular lipid content was observed using Oil Red O staining,and intracellular TG,malondialdehyde (MDA ),SOD and reactive oxygen species (ROS)levels were determined by the assay kits. The protein expression of nuclear factor erythroid 2-related factor 2 (Nrf2),heme oxygenase-1 (HO-1 )and NAD (P)H quinone oxidoreductase 1 (NQO-1 )were measured by Western blot. The results showed that TUA significantly increased SOD activity,and decreased intracellular TG, ROS and MDA levels in FFAs-induced HepG2 cells. Moreover,TUA dramatically improved Nrf2,NQO-1 ,and HO-1 expression. However,the dramatic increase in TG,ROS,MDA levels and the reduction in SOD,NQO-1 and HO-1 expression following Nrf2 inhibitor brusatol treatment were observed. In conclusion,these results suggest that TUA has the therapeutic effect on NAFLD which may be associated with Nrf2 activation.

A ingestão de bebidas alcoólicas é um evento socioculturalmente aceito em muitos países. Porém, o consumo frequente e descontrolado deste tipo de bebida configura o transtorno por uso de álcool (TUA). Esta condição causa agravos que podem afetar a sociedade de uma forma geral. O TUA também pode levar os pacientes a contraírem doenças. Entre estas, existe uma relação importante entre TUA e doenças infectocontagiosas, com destaque para a infecção pelo HIV e o posterior desenvolvimento da AIDS. Portanto, a presente pesquisa objetivou realizar uma revisão da literatura sobre as relações entre TUA e HIV/AIDS. A seleção do material científico foi efetuada tendo por base plataformas eletrônicas, tais como: Google Scholar, MEDLINE, LILACS, SciELO, NCBI / PUBMED, Scopus e Science Direct. O entendimento dos fatores relacionados ao TUA, principalmente em pacientes com HIV/AIDS, é de fundamental importância para a formulação e criação de estratégias de políticas públicas que visem reduzir esta possível relação (AU)


The ingestion of alcoholic beverages is socio-culturally accepted in many countries. However, frequent and uncontrolled consumption of this type of beverage constitutes alcohol use disorder (AUD). This condition may be harmful to society in general, and it can lead patients to contract other diseases. There is an important relationship between AUD and infectious diseases, with emphasis on HIV infection and the later development of AIDS. Therefore, the present research aimed to carry out a review of the literature on the relationship between AUD and HIV/AIDS. The selection of the scientific material was based on electronic platforms, such as Google Scholar, MEDLINE, LILACS, SciELO, NCBI/ PUBMED, Scopus and Science Direct. The understanding of the factors related to AUD, especially in patients with HIV/AIDS, is of fundamental importance for the formulation and creation of public policy strategies aimed at reducing this possible relationship (AU)

ABSTRACT

INTRODUÇÃO: A espécie vegetal Jatropha gossypiifolia L., conhecida também como pião roxo, é utilizada na medicina popular como cicatrizante, anti-hipertensivo, purgativo e diurético. OBJETIVO: Avaliar a influência da administração intraperitoneal da Jatropha gossypiifolia L., na cicatrização de suturas da parede abdominal ventral de ratos, observando-se os seus aspectos macroscópicos, tensiométricos e microscópicos. MÉTODOS: Foram utilizados no procedimento 40 ratos da linhagem Wistar, machos, distribuídos em dois grupos de 20. Após incisão da parede e abertura da cavidade abdominal, foi instilado 1 ml/kg/peso de cloreto de sódio a 0,9 por cento no grupo controle e no grupo Jatropha o extrato bruto etanólico da Jatropha gossypiifolia L., na concentração de 1 ml/kg/ peso. Realizou-se a sutura da parede abdominal com fio de polipropileno, com pontos separados. Os animais foram avaliados na sua evolução pós-operatória e mortos em dois subgrupos, no 3° e 7° dia. Analisou-se a parede abdominal ventral macroscopicamente, mediu-se a força de resistência a tensão e foram estudados os aspectos histológicos do reparo cicatricial. RESULTADOS: No exame macroscópico encontraram-se aderências mais intensas nos subgrupos Jatropha no 3° e 7° dia. A avaliação tensiométrica foi significantemente maior nos subgrupos Jatropha no 3° e 7° dia. A avaliação histológica comparativa entre os subgrupos demonstrou que o processo inflamatório agudo foi significantemente maior no subgrupo Jatropha no 3° e 7° dia; a neoformação capilar foi significantemente maior no 3° dia pós-operatório do subgrupo Jatropha sendo os outros parâmetros histológicos semelhantes. CONCLUSÃO: O uso do extrato bruto de Jatropha gossypiifolia L. intraperitoneal não demonstrou melhora significativa no processo de cicatrização da sutura da parede abdominal ventral de ratos com a dose e concentração utilizadas.


INTRODUCTION: The Jatropha gossypiifolia L., which is used in popular medicine is considered to have good diuretic effect in hypertension and is also used as a laxative drug. It seems to have a healing effect, although not proved till now. PURPOSE: To evaluate the influence of intraperitoneum administration of Jatropha Gossypiifolia L., in suture healing of ventral abdominal wall of rats, through tensiometric measurement, macro and microscopic aspect of post-operative period. METHODS: Forty wistar male rates were allocated in two groups of 20 animals . After the incision and exposure of abdominal cavity 1 ml/kg/weight of 0,9 percent sodium chloride solution was injected in control group, and in the other one the injection was of 1 ml/kg/weight of a gross ethanol extract of Jatropha gossypiifolia L. The suture of the abdominal wall was than performed with polypropylene separated stitches. The animals were followed-up and killed in the third and seventh days. The ventral abdominal wall was macroscopically analyzed, the resistance strength to strain was measured and it was also studied the histological aspects. RESULTS: On macroscopic examination more intense adhesion was found on the group of Jatropha in both third and seventh post-operative days. The strain evaluation was meanly greater on Jatropha group also in third and seventh days. CONCLUSION: The histological comparative analysis between the different groups showed that the acute inflammatory process was meanly greater for the Jatropha group in third and seventh post-operative days. The vascular neoformation was significantly greater in third pos-operative day of Jathopha group; the other histological parameters were just alike. The intraperitoneum injection of Jatropha extract did not have any significant improvement for the wound healing on ventral abdominal wall on the evaluated animals in this study, no matter if analyzed at the third or seventh pos-operative days.
